Zij werd bekend als zangeres bij het dansorkest van Les Brown. Tot haar meest bekende liedjes behoren ‘It's magic’ (Romance on the high seas, 1948), ‘Secret love’ (Calamity Jane, 1953) en ‘Que sera, sera’ (in Hitchcocks The man who knew too much, 1956). Nadat ze succes had geoogst in musicals en thrillers, verwierf ze haar grootste populariteit met licht pikante liefdeskomedies als Pillow talk (1959), die haar jarenlang tot Hollywoods grootste kassamagneet maakten. Daarna had ze van 1968 tot 1973 haar eigen tv-show "The Doris Day Show", alvorens ze zich geheel ging wijden aan de dierenbescherming. Haar memoires publiceerde zij in Doris Day: her own story (1975).
